Multilayer films composed of anionic, cationic, and zwitterionic polymers have been prepared by electrostatic interaction between oppositely charged polyelectrolytes in aqueous solutions. The polyelectrolyte systems have been studied with respect to pH-dependent electrostatic interactions that result in control over the internal structure of multilayers and the delivery of different cargoes, including drugs, proteins, and gene-silencing agents. The antifungal drug amphotericin B was incorporated within the layers of the multilayers, which increased the antifungal activity of the drug. In contrast, the incorporation of dextran into multilayers of poly(allylamine) and poly(styrenesulfonate) prevented the swelling of the multilayers. The multilayer structure itself was stable over a wide range of pH (3.0-12.0) and ionic strength (0-300 mM). The multilayer films also prevented the diffusion of small molecules such as dextrans, quinacrine, and carboxyfluorescein. The release of anionic, cationic, and zwitterionic poly(allylamine) multilayers loaded with the hydrophobic anticancer drug paclitaxel was pH-dependent. The cationic and zwitterionic systems released more of the drug at lower pH, whereas the anionic systems did not release significant amounts of drug until the pH was elevated to 4.0. The poly(allylamine) system was also stable against DNase I digestion. Furthermore, two different genes, cotransfected into a human adenovirus vector, were silenced within the layers of poly(allylamine) and poly(styrenesulfonate) multilayers prepared from different cationic polyelectrolytes.Q:
